In Buffalo, the business loop follows former U.S. 87 southbound and merges with U.S. 16 eastbound, avoiding downtown Buffalo entirely. This business loop is cosigned with Business Loop I-25 along the north-south section and with U.S. 16 on the east-west section.

After departing from Interstate 90, the business route follows Main Street south from the freeway toward downtown Buffalo, crossing the city limits almost immediately. The first shield assembly on southbound (eastbound) Business Loop I-25, Business Loop I-90, and Business U.S. 87 features a "TO" banner above the Business Loop I-25 shield, even though the guide sign at the exit ramp from eastbound Interstate 90 does not indicate "TO." This further complicates where exactly where the northern terminus of Business Loop I-25 is ... and the best answer is that it has three possible end points: in downtown Buffalo at the northern U.S. 16 intersection, the Interstate 25 interchange with U.S. 16 (Exit 299), and the Business Loop I-25/Business Loop I-90 interchange with Interstate 90. Southbound Business Loop I-25, Business Loop I-90, and Business U.S. 87 approach Airport Road, which travels northwest to the Johnson County/Buffalo Airport. About six blocks south of there, Business Loop I-25, Business Loop I-90, and Business U.S. 87 approach Junction U.S. 16 (Hart Street) east, which connects to both Interstate 25 and Interstate 90 before meeting U.S. 14 near Ucross. Reaching Business Loop I-25 and Business U.S. 87 is this somewhat confusing sign. It correctly states that a turn in either direction will connect to Business U.S. 87, and the sign shows U.S. 16 turning south (left) at this intersection. However, the I-90 Business Loop is shown to turn left, toward downtown, even though Business Loop I-90 actually turns right here, as if to return back to its parent route.
